John Stanley was born in England and brought over in 1634 by his father, who died on the passage. By order of the General Court of 3 March 1634/5 he was put with his uncle Thomas Stanley, who took fifty eight pounds of the estate to bring him up and was to pay him fifty pounds at twenty one. He was about ten.

He settled at Farmington, Connecticut. He married first, on 5 or 15 December 1645, Sarah, daughter of Thomas Scott of Hartford, who bore him six children and died on 26 June 1661, and second, on 20 April 1663, Sarah, daughter of John Fletcher of Milford, by whom he had two daughters. He was made freeman in 1665, served as Lieutenant and afterwards Captain up the Connecticut river through King Philip's War, and sat as representative. He died on 19 December 1705, having made his will the April before.

His daughter Sarah married Joseph Gaylord of Windsor on 14 July 1670 and went with him to Mattatuck and then to Durham. His son Timothy settled at Waterbury, commanded its train band through the Indian war, sat as deputy to the General Court nine times, and died childless on 12 November 1728. Timothy's will names his heirs: Timothy, Samuel, Nathaniel and John Stanley, sons of his brother John, and Joseph, John and Benjamin Gaylord, Ruth Hickox and Johannah Royce, the children of his sister Sarah Gaylord.
