John Tilley was baptized at Henlow, in Bedfordshire, on 19 December 1571, "Jn s Rob TILLYE." His father, Robert Tilley, was a Henlow yeoman. His younger brother Edward, baptized in 1588, also sailed on the Mayflower.

On 20 September 1596 he married Joan Rogers at Henlow. She was born Joan Hurst, the daughter of William Hurst of Henlow, and was the widow of Thomas Rogers. The register calls John a husbandman. Their children were baptized at Henlow: Rose in 1597, who died young, John in 1599, a second Rose in 1602, Robert in 1604 and Elizabeth on 30 August 1607. His father's will of 1612 provided for "John Tilleye and his children."

John Tilley came to Plymouth on the Mayflower in 1620 with his wife and their daughter Elizabeth. Edward Tilley, his wife Ann and two young cousins of theirs, Henry Samson and Humility Cooper, came on the same ship.

In December 1620 John Tilley was one of the eighteen men who took the shallop along the shore of Cape Cod Bay to find a place to settle. Myles Standish, John Carver, William Bradford, Edward Winslow, Edward Tilley and John Howland were with him. On 8 December they were attacked by the Nauset at a place now known as Eastham. They spent the next days on Clark's Island and landed at Plymouth on 11 December.

He did not survive the first winter. Bradford wrote that "John Tillie and his wife both dyed a litle after they came ashore; and their daughter Elizabeth maried with John Howland, and hath issue as is before noted." His wife's name is not in Bradford. It was Joan, as Elizabeth's baptism shows.
