Anna Marie Grauer was born at Kusterdingen, in Württemberg, on 2 October 1844, and was baptised there the next day as Anna Maria. Kusterdingen is a village a few miles east of Tübingen. Her parents were Jacob Grauer and Anna Maria Vött, who had married in the village in 1837. Her husband, Nicholas Wilhelm Ruppert, was a Saxon, so the couple met across old German lines. She raised at least eight children on the Nokomis farm.

She was the fourth of at least six children baptised at Kusterdingen: Jakob in 1838, a second Jakob in 1840, Wilhelm in 1842, herself in 1844, Conrad in 1847 and Agnes in 1849. Her brother Jacob farmed at Marysville in Kansas and died there in 1901. Her brother Conrad farmed at Palmer, in Christian County, Illinois. When Conrad died in July 1917, his obituary in the Decatur Review listed his children and ended: "One sister, Mrs. Nicholas Ruppert, of Nokomis, also survives." Another Kusterdingen Grauer, George, born in 1835, also farmed at Marysville. He was born two years before her parents married, and how he was related to her is not known.

Her son's wife had a mother from the same village. Anna Gutbrod, baptised at Kusterdingen in May 1845, less than eight months after her, married Jacob Zimmerman in Logan County, and their daughter Regena married Anna Marie's son John in 1889. Anna Gutbrod's own sister married a Grauer at Marysville. So both of John and Regena Ruppert's mothers were born in one Württemberg village.

Widowed in November 1917, four months after her brother Conrad died, she moved into John's household on Spruce Street, where the 1920 census finds her at seventy-five. She died there on 20 March 1924. She shares one stone with her husband at Saint Paul's Cemetery, Nokomis. It reads "ANNA MARIE RUPPERT" and carries a German couplet: "Nach überstand'nen schweren Leiden / Bin ich versetzt in hohen Freuden", after the heavy sorrows overcome, I am carried into high joys.
