Increase Niles was born at Braintree on the sixteenth of the tenth month, 1646, the sixth of the seven children entered in the town register to John Niles and his wife Jane.

On 4 December 1677 he married Mary Purchase, the ceremony performed by the governor of the colony, John Leverett. Their son John was born at Braintree the following October, and the clerk entered him as 'John Nile, the son of Increase, Nile & Mary his wife was borne ye 10th October 1678'. The town wrote the name Nile as often as Niles.

In December 1675, during King Philip's War, he mustered at Dedham in the Braintree file of Captain Isaac Johnson's company, raised for the expedition against the Narragansett. Nine days later the company went into the Great Swamp Fight, where Captain Johnson was killed. The list does not say whether he was in the fight.

A second son, Increase, was born on 9 March 1680/1. Joseph W. Porter says he and his wife were original members of the second church of Braintree. He died on 1 September 1693, entered by the clerk as "Incres Niles." Four years later his widow married Francis Nash, the man written beside him on the 1675 muster, and his son John married Nash's daughter Margaret.
