David Cox was born about 1746, a son of Isaac Cox and Susannah Tomlinson. He grew up on the South Branch of the Potomac in Hampshire County, Virginia. There, about 1768, he married Margaret Bruce.

In the early 1770s the Coxes moved to the upper Ohio, into the country Virginia then claimed as Yohogania County and which later became southwestern Pennsylvania. David served there as a lieutenant of militia and as a constable from 1778. His brother Isaac sat on the county court. On 27 September 1779 the court recorded a power of attorney from David Cox to Friend Cox, a kinsman of the same name his own son carried.

In March 1780 David took his family down the Ohio to Kentucky with his brothers Isaac and Benjamin. Indians attacked the party on the way and killed his brother Joseph. On 22 May 1780 the Yohogania court named Jeremiah Wright lieutenant of militia "in the stead of David Cox who has removed out of the County." The brothers built Cox's Station on the middle branch of Cox's Creek, five miles northeast of the present Bardstown. When Nelson County was organized in 1785, David and his brother Isaac were among its first justices.

Isaac died in 1788, killed by Indians. His will left five hundred pounds to David's sons Friend, David and James.

David and Margaret had eleven children: James, Jane, Friend, Mary, David, Margaret, Susannah, Isaac, Jonathan, Gabriel and George. Their daughter Margaret, called Peggy, married her cousin Friend McMahon in January 1798, the son of David's sister Ann. Margaret Bruce Cox died before 1808. On 28 July 1808 David married Susanna Martin in Bullitt County.

He made his will on 14 February 1818. He died in Nelson County on 18 November 1820, aged about seventy-four, and the will was proved on 18 December. He is buried in the churchyard of Cox's Creek Baptist Church, in the community his family founded.
