Mary Jane Mullins was born on 15 May 1815 in North Carolina, a daughter of John Mullins and his wife Ollie. When she died in 1893 the Dickenson County death register gave her birthplace as Yancey County, North Carolina, the name the county carried from 1833.

In 1829, when she was fourteen, she came with her father, John Mullins, and three younger brothers to Holly Creek in southwest Virginia, where the town of Clintwood now stands. More than a century and a half later a local history group, Mountain People and Places, set a granite monument to her in front of the Jonnie B. Deel Library in Clintwood. It is headed "Pioneer Lady, 1815, 1893" and reads: "In 1829 Mary Jane Mullins, at age fourteen, was the first pioneer lady to settle here at Holly Creek (now Clintwood) with her father John Mullins and three younger brothers. Four years later she married John Jack Fleming and raised twelve children. For sixty four years until her death she made this area home."

She married John J. Fleming about 1833, at seventeen or eighteen. Family accounts say the couple settled first on Beaver Creek in Floyd County, Kentucky, and their first three children were born in Kentucky: John in August 1834, Lavenia and Isaac. By about 1841 they were back on Holly Creek, at the farm that became Flemingtown.

In 1850 the census taker wrote her as Mary Fleming, 34, born in North Carolina, with her husband John and eight children at home: John, Lavenia, Isaac, Emmanuel, Martha, Sally, William and David. Robert, George W., Mary Jane and Andrew J. came after. When her son William married Martha J. Branham in Wise County in 1874, the clerk entered his parents as John and Mary Fleming.

John died in February 1881. Mary lived on at Flemingtown for twelve more years and died there on 6 October 1893, aged 78. The county register entered her as Mary Fleming, a married woman of 79, a seamstress, and named her parents John Mullins and Ollie. She was buried beside him in the Fleming Cemetery. Her stone reads "Mary J., wife of John J. Fleming, Born May 15, 1815, Died Oct. 6, 1893," and below, "A tender mother and faithful friend."
