Moses Tyler was born at Andover or Roxbury, Massachusetts, in 1641 or 1642. He was a son of Job Tyler and his wife Mary. His father had been at Newport, Rhode Island, in 1638 and was among the first at Andover. In June 1659 Moses swore to a deposition with his parents and his sister Mary, and the record gives his age as "between 17 and 18 yeares". That line is how his birth is dated.

On 6 July 1666 he married Prudence Blake, daughter of George and Dorothy Blake of Gloucester, "early settlers" of Boxford in the words of Sidney Perley's History of Boxford (1880). Prudence was born on 15 April 1647. Perley lists eight sons: Moses on 16 February 1667, John on 14 September 1669, Joseph on 18 September 1671, Ebenezer on 17 December 1673, Job on 16 December 1675, Samuel on 2 May 1678, Nathaniel on 14 August 1680 and Joshua on 4 July 1688. Job was born in Rowley Village. Prudence died on 9 March 1689.

Moses served Boxford as selectman, committee-man and surveyor, and he was known as "Quartermaster" Tyler. A quartermaster was the officer of a militia troop who saw to its supplies and quarters.

He married a second wife, Martha, who was born in 1649. Perley says he was living in 1712. He died at Andover on 2 October 1727, aged about eighty-five, by the Tyler genealogy of 1912. His will was proved at Salem on 13 October 1727. It named his son Job among seven sons who were given their portions. Martha outlived him and died on 13 February 1735, aged eighty-six.
