Samuel Gurley was born at Northampton, Massachusetts, on 17 May 1687, the only child of William Gurley and Hester Ingersoll. His father had drowned in the Connecticut River sixteen days before.

When he was not quite two, his mother married Benoni Jones of Northampton, on 23 January 1689. Four half-brothers followed: Jonathan in 1695, who died young; Benjamin in 1696; Ebenezer in 1698; and a second Jonathan in 1703.

On 13 May 1704, when Samuel was sixteen, French soldiers and their Native allies took the outlying garrison at Pascommuck, at the north end of Mount Tom. His stepfather and his half-brothers Ebenezer and Jonathan were killed. His mother was carried to Canada, where she died on 27 November 1705. Where Samuel was that day is not recorded.

About 1712 he married Experience Rust, a daughter of Nathaniel Rust of North Coventry, Connecticut, who had come there from Northampton. His half-brother Benjamin Jones also settled at Coventry. Samuel and Experience lived first at North Coventry and then at Mansfield. They had eleven children between 1713 and 1731.

The family history of 1897 says he was "a farmer, and lived near the old Gurley burying-ground" and was "a prominent man of the town". He died on 23 February 1760. His record reads: "Mr. Samuel Gurley died February 23, 1760, in the 74th year of his age. 'He was distinguished for piety, and was eminently useful in the cause of religion and humanity.'" His widow lived until July 1768.
