Sarah Horton was born about 1598, probably in the London parish of St Martin Vintry. Her father, Thomas Horton, was a citizen and mercer of London. Her mother, Catherine Satchfield, was his second wife. She had a younger sister, Parnell, and several older half-brothers and half-sisters from her father's first marriage. Her father made his will in 1615 and died before January 1621.

She married Roger Conant, a London salter, at St Ann's, Blackfriars, on 11 November 1618. Their first child, Sarah, was christened at St Lawrence Jewry in September 1619 and buried there in October 1620. A son, Caleb, was christened there in May 1622.

She crossed to New England with her husband and Caleb in 1623 or 1624. She lived with him at Plymouth, at Nantasket, at the Cape Ann fishing station, and from the fall of 1626 at Naumkeag, which became Salem. Her son Lot was born about 1624. Her son Roger, born in 1626, was called the first white child born in Salem. Then came Sarah, Joshua, Mary, Elizabeth and, in 1637, Exercise. About 1640 the family numbered nine, as Roger wrote in the Salem book of grants.

Caleb went back to England. His grandmother Catherine Horton named him in her will of 1627, and he died there unmarried before November 1633.

She was living in 1666, when a deed names her. She signed with her husband and sons as a member of the new church at Beverly in June 1667, and she is missing from a list of the same people that September. She probably died that summer. She is not named in her husband's will of 1678.
