Elizabeth Scott was baptised at Rattlesden, in Suffolk, England, on 18 November 1623. She was the eldest child of Thomas Scott, a glover, and his wife Elizabeth Strutt. Her grandparents were Henry Scott and Martha Whatlock of Rattlesden. Walter Goodwin Davis's Ancestry of Phoebe Tilton (1947) gives her as "ELIZABETH, bapt. in Rattlesden Nov. 18, 1623; m. John Spofford of Rowley."

She was ten when her father sailed for New England in the Elizabeth of Ipswich in April 1634, with his widowed mother and his sister Ursula Kimball's family. Her grandmother brought ten grandchildren with her, and Elizabeth was very probably one of them. Her father settled at Ipswich, Massachusetts.

She married John Spofford, who had come to Rowley with the Rev. Ezekiel Rogers's company from Yorkshire. Their first child, Elizabeth, was born on 14 December 1646. John followed on 24 October 1648, Thomas in 1650, Samuel in 1653 and Hannah in 1654. Mary was born in 1656. A daughter Sarah, born in January 1658, died at two, and a second Sarah was born in 1662. Francis, the youngest, was born in 1665.

In the spring of 1669 the family moved out to a farm the town leased to her husband, on what is still called Spofford's Hill, in the present Georgetown. Her father had died at Ipswich in 1654, leaving her £25.

Her husband made his will in October 1678. To "my dear and loving wife" he left the lease of "the house and lands of Mrs. Prudence Cotton", all the household stuff "except the arms and ammunition", two cows, a calf and four sheep, and the use of four acres at the farm for her life. Francis, then thirteen, was to stay under her direction until he was twenty-one, "that he may be helpfull to her to carry on her husbandry work." The will does not give the wife's name. Jeremiah Spofford's genealogy of 1888 takes her to be Elizabeth. Her husband died that autumn. Of Elizabeth's own death the same book says only, "We have no record of her death."
