Isaiah Huntley was born at Lyme, Connecticut, on 12 November 1752, according to a family genealogy printed in 1914. His gravestone gives his age at death as sixty-nine, which points to 1751. A Huntley genealogy printed in 1978 records him as "Isaiah Huntley (Nathan, Aaron, Aaron, John) born in Lyme, Conn." His father was Nathan Huntley and his mother Lucy Smith, both of Lyme. Behind Nathan the line ran through two Aarons to John Huntley, who lived at Boston and Roxbury before settling at Lyme.

Three months after Isaiah was born, on 17 February 1753, his father bought family land at Lyme.

In the Revolution he served as a private in Colonel Bellows' regiment of militia. In June 1777 that regiment marched to reinforce the garrison at Fort Ticonderoga, the stone fort at the narrows of Lake Champlain. Bellows' was a New Hampshire regiment. Within weeks a British army under General Burgoyne came down the lake from Canada, and the garrison abandoned the fort in the first days of July. The 1978 genealogy gives a second account, that he "served in the American Revolution in Capt. Canfield's" company. A lineage book of the Daughters of the American Revolution names the captain as Samuel Canfield.

On 5 September 1779 he married Elizabeth Church, daughter of Simeon Church, the blacksmith of Chester parish in Saybrook. Their seven children were born at Marlow, New Hampshire: Calvin in August 1780, Nathan in January 1782, Isaiah in November 1787, Leland in December 1790, Allen in December 1792, Welthy in March 1795 and Selden in October 1797. The family was still at Marlow in 1803, when Elizabeth was listed among her father's heirs as "Eliza, wife of Isaiah Huntley, Marlow, N.H."

The family later moved on into Vermont. Isaiah died on 21 December 1820, aged sixty-nine by his stone. He and Elizabeth are buried in the Duxbury Corner Cemetery, which the genealogy places at Waterbury, Vermont.
