Mehitable Scotto was baptized at the First Church of Boston on 11 February 1649, "Mehetabell Scottoe the Daughter of our brother Thomas Scottoe being about 4 Dayes old." Her parents were Thomas Scotto and his first wife Joan. Two of her older brothers, both named Thomas, had died at birth. A brother John was born in 1644 and another Thomas in 1647. Her mother died before 1655, and her father married again. Her grandmother Thomasine, who had brought the family from Great Yarmouth in 1634, was still alive in 1660.

Her father died in 1661, when she was twelve. His will left equal shares to all his children. Her uncle Joshua Scotto was a founder of the South Church and later a judge in Maine.

In 1671 she married Andrew Clarke, son of Thomas Clarke of Plymouth. They lived near Scottow's Lane, the street that carried her family's name. Her children were Thomas, Susanna, Andrew, Scotto, Nathaniel and Mehitable, born between 1672 and 1686. Her son Scotto carried her maiden name, and it passed down through three generations of Harwich Clarks.

In 1694 the family moved to Harwich, on Cape Cod. Her husband died there in 1706. She died on 24 April 1712 and was buried beside him in the old burying ground of Harwich's north parish, now Brewster. Her stone reads: "Mehitable Clarke, widow of Andrew Clarke, died April ye 24th 1712, aged about 62 years."
