Sarah Spencer was born in 1635, a daughter of William Spencer of Hartford, Connecticut, and his wife Agnes. Her father made his will on 4 May 1640 and died that year, when she was about five. He left a third of his estate "to my two daughters, Sarah and Elizabeth," and asked that the children be "brought vppe wth the improvement of the whole estate." Her mother married again, to William Edwards of Hartford.

When the court settled her father's estate in 1650, a bond secured five pounds "unto Sara Spencer, at the age of eighteen yeares, wch will be in the yeare 1653." That fixes her birth in 1635. If all the children died before their portions came due, the money was to go to "Agnes Edwards ... the mother of the said children."

She married John Case and lived with him at Windsor. Their eldest child, Elizabeth, was born about 1658. Mary followed on 22 June 1660, John on 5 November 1662, William on 5 June 1665 and Samuel on 1 June 1667. In the spring of 1669 the family moved up to Simsbury and settled at Weatogue, on the Farmington River. Her husband was made the first constable of Simsbury on 14 October 1669. Richard was born on 27 August 1669, Bartholomew in October 1670, Joseph on 6 April 1674, Sarah on 14 August 1676 and Abigail on 14 May 1682. She bore ten children in twenty four years.

Her brother Samuel stayed at Hartford. Years after her death her husband named "my brother Samuel Spencer of Hartford" an overseer of his will.

She died at Simsbury on 3 November 1691, aged fifty five. Her husband married again, to Elizabeth, widow of Nathaniel Loomis of Windsor. Her headstone survives in the Simsbury cemetery, a small arched stone of red sandstone.
